Stable isotopes in NMR-spectroscopy

 

Nuclear magnetic resonance (NMR)-spectroscopy is used for 3D structure and dynamic clarifications. For this purpose, the molecule of interest gets labelled with the nuclear spin active stable isotopes 13C and 15N. Silantes focusses on NMR analyses of nucleic acids (RNA, DNA) and proteins. These compounds are produced with a biotechnological approach using microorganisms such as bacteria, yeast and algae in a biotechnological approach.

 

Stable isotopes in Mass spectrometry

 

Proteins are the effectors of all activities within cells. The expression of certain proteins change upon cell stimulation or stress, or when cells differentiate or turn into a disease state. The measurement of protein expression levels as well as the  characterization of their post-translational modifications gives information about their cellular state. The goal of quantitative proteomics is to evaluate the relative expression of proteins between two cellular samples being compared.

 

Mass spectrometry (MS) is used for the quantification and identification of molecules.  For the accurate quantification of proteins, the stable isotope labelling of amino acids in cell culture (SILAC) is a versatile tool. For this purpose, reference molecules differing from the sample molecule with regards to isotopic labelling (2H, 13C, 15N) are necessary.
